Blocking and diluting buffer and concentration : 5% NFDM/TBST

Lysates were kindly provided by collaborator.

ab236629 Anti-PSRC2 antibody [EPR20913-32] was shown to react with PSRC2 in mouse lymph node tissue lysate in Western blot. Loss of signal was observed when PSRC2 knockout sample was used. Wild-type and PSRC2 knockout samples were subjected to SDS-PAGE. ab236629 and ab129002 (Rabbit anti-Vinculin loading control) were incubated 1 hour at room temperature at 1/1000 dilution and 1/5000 dilution respectively. Blots were developed with Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated (ab97051) secondary antibody at 1/100, 000 dilution for 1 hour at room temperature before imaging. he blot was developed on a BIO-RAD® ChemiDoc™ MP instrument using the ECL technique.

PSRC2 also known as Proline/Serine-rich Coiled-coil 2 is a protein with a known mass of approximately 28 kDa. This protein is expressed in a variety of tissues with notable presence in the brain and testis. Mechanically PSRC2 participates in cellular processes through its role in cytoskeleton organization. It exhibits specific binding properties which facilitate its involvement in microtubule dynamics aiding in the stabilization or destabilization of these structures during cell division.
