Supplementary information

PSS-2 also known as Phospholipid Scramblase 2 functions mechanically as a lipid scramblase enzyme. It mediates the bidirectional movement of phospholipids across the bilayer of cellular membranes facilitating the collapse of lipid asymmetry. PSS-2 has a molecular weight of approximately 37 kDa. It is expressed in various tissues with significant presence in the liver which indicates its importance for lipid metabolism in this organ.
Biological function summary

Phospholipid scramblase 2 contributes to lipid homeostasis and membrane dynamics. PSS-2 forms complexes with several membrane proteins playing a role in processes such as apoptosis and cell signaling. By facilitating phospholipid translocation PSS-2 helps maintain cellular membrane composition which is critical for membrane integrity and function. This activity impacts cellular processes including cell shape motility and interaction with the environment.

Pathways

The activity of phospholipid scramblase 2 integrates into lipid metabolism and apoptotic pathways. Within the lipid metabolism pathway it cooperates closely with fatty acid-binding proteins to regulate lipid distribution and metabolism. Its involvement in the apoptotic pathway partners with caspases where it contributes to phosphatidylserine externalization an important apoptosis signal. This coordination places PSS-2 as a significant player in maintaining cellular health.

Dysregulation of phospholipid scramblase 2 associates with liver disorders and cardiovascular disease. Alterations in PSS-2 expression relate to non-alcoholic fatty liver disease due to its central role in hepatic lipid metabolism. Furthermore the interaction with proteins such as annexins links PSS-2 to cardiovascular health as these interactions influence thrombosis and inflammation. Understanding PSS-2's contributions in these contexts could lead to potential therapeutic targets for managing these conditions.

Target data

Catalyzes a base-exchange reaction in which the polar head group of phosphatidylethanolamine (PE) or phosphatidylcholine (PC) is replaced by L-serine (PubMed : 19014349). Catalyzes the conversion of phosphatatidylethanolamine and does not act on phosphatidylcholine (PubMed : 19014349). Can utilize both phosphatidylethanolamine (PE) plasmalogen and diacyl PE as substrate and the latter is six times better utilized, indicating the importance of an ester linkage at the sn-1 position (By similarity). Although it shows no sn-1 fatty acyl preference, exhibits significant preference towards docosahexaenoic acid (22 : 6n-3) compared with 18 : 1 or 20 : 4 at the sn-2 position (By similarity).
