PTF1A was immunoprecipitated from 1 mg of NIH/3T3 (Human epithelial cell line from cervix adenocarcinoma) whole cell lysate with ab182398 at 1/50 dilution. Western blot was performed from the immunoprecipitate using ab182398 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.

Lane 1 : NIH/3T3 whole cell lysate 10 μg (Input).

Lane 2 : ab182398 IP in NIH/3T3 whole cell lysate.

Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab182398 in NIH/3T3 whole cell lysate.

PTF1A also known as pancreas specific transcription factor 1a or PTFL is a bHLH (basic Helix-Loop-Helix) transcription factor with a mass of approximately 35 kDa. This protein functions as a DNA-binding protein that regulates gene expression. It is mainly expressed in the pancreas especially within pancreatic progenitor cells and also in certain neural tissues. PTF1A is necessary for the transcriptional activity of genes involved in pancreatic development.
Biological function summary

The PTF1A protein plays a significant role in pancreas development and maintenance. It forms a complex as part of the pancreas transcription factor 1 (PTF1) complex which includes other proteins like RBPJL and the E-box binding protein. This complex binds to specific regulatory regions of DNA to modulate the expression of genes important for acinar cell differentiation and function in the pancreas.

Pathways

PTF1A operates within the Notch signaling pathway and the Wnt signaling pathway. In the Notch pathway PTF1A interacts with proteins such as Notch receptors and target HES genes to influence cell differentiation. Within the Wnt pathway PTF1A impacts genes involved with cell fate decisions during pancreatic development working with signaling proteins like beta-catenin to mediate its effects.

PTF1A mutations are associated with pancreatic agenesis and cerebellar atrophy. Pancreatic agenesis involves the underdevelopment or absence of the pancreas resulting in glucose homeostasis complications. PTF1A can interact with genes such as PDX1 in this context which is another transcription factor involved in pancreatic development. In cerebellar atrophy the disruptions in PTF1A expression can lead to problems with cerebellar development causing neurological symptoms due to its interaction with neural development pathways.

Plays a role in early and late pancreas development and differentiation. Important for determining whether cells allocated to the pancreatic buds continue towards pancreatic organogenesis or revert back to duodenal fates. May be involved in the maintenance of exocrine pancreas-specific gene expression including ELA1 and amylase. Required for the formation of pancreatic acinar and ductal cells. Plays an important role in cerebellar development.
