Supplementary information

Prothymosin alpha (PTMA) also known as prothymosin is a small nuclear protein characterized by a polypeptide chain with a molecular mass of approximately 12 kDa. PTMA is broadly expressed across various tissues with abundant levels in the thymus and spleen. Its sequence is rich in acidic amino acids a feature that influences its interaction with other biomolecules. Prothymosin alpha undergoes post-translational modifications further affecting its functions within the cell.
Biological function summary

Prothymosin alpha plays a role in chromatin remodeling and regulation of gene expression. It acts as part of the chromatin-modifying complex interacting with other proteins to influence transcription processes. PTMA contributes to cell proliferation and apoptosis aiding cellular defense mechanisms. Its presence in the nucleus is significant for the cell cycle where it regulates the transition between phases.

Pathways

PTMA participates in the apoptotic pathways and the regulation of gene transcription. It works closely with proteins such as histone acetyltransferase complexes which are important for modifying chromatin structure and facilitating transcription. Prothymosin alpha interacts with apoptosis regulatory proteins impacting how cells respond to apoptotic signals and controlling cellular growth and death.

PTMA has connections with cancer and autoimmune diseases. Its involvement in cell proliferation links it to oncogenesis where overexpression of PTMA has been observed. The protein also interfaces with immune regulatory proteins influencing autoimmune conditions. PTMA’s dysregulation can induce abnormal cell cycle progression contributing to disease development.
