PTPMT1 also known as protein tyrosine phosphatase mitochondrial 1 has a molecular mass of approximately 23 kDa. This protein is a unique phosphatase localized to the mitochondria. It specifically dephosphorylates phosphatidylinositol phosphates and plays a critical role in mitochondrial function. PTPMT1 is expressed highly in tissues with significant energy demands like the heart and skeletal muscle suggesting its role in energy metabolism.
Biological function summary

A phosphatase that is involved in important tasks within the mitochondria PTPMT1 contributes significantly to the regulation of mitochondrial metabolism. It removes phosphate groups from target substrates which is essential for the biosynthesis of cardiolipin an important lipid in mitochondrial membrane composition. Although not formally part of a large complex it interacts closely with other mitochondrial enzymes to maintain lipid homeostasis.

Pathways

Lipid metabolism and insulin signaling both involve PTPMT1. It mediates its actions through the biosynthesis of phosphatidylinositol derivatives that influence the insulin signaling pathway. It has a connection with the PI3K/Akt signaling cascade affecting the actions of proteins such as Akt and PDK1 which play integral roles in cellular energy balance and glucose homeostasis.

Improper PTPMT1 function relates closely to metabolic disorders particularly diabetes mellitus. PTPMT1 activity affects insulin signaling and its dysregulation contributes to insulin resistance. There is also an association with mitochondrial dysfunction which may lead to neurodegenerative disorders. In these contexts PTPMT1's interaction with proteins like insulin receptor substrate (IRS) can provide avenues to study therapeutic targets for related diseases.

Lipid phosphatase which dephosphorylates phosphatidylglycerophosphate (PGP) to phosphatidylglycerol (PG) (By similarity). PGP is an essential intermediate in the biosynthetic pathway of cardiolipin, a mitochondrial-specific phospholipid regulating the membrane integrity and activities of the organelle (By similarity). Has also been shown to display phosphatase activity toward phosphoprotein substrates, specifically mediates dephosphorylation of mitochondrial proteins, thereby playing an essential role in ATP production (By similarity). Has probably a preference for proteins phosphorylated on Ser and/or Thr residues compared to proteins phosphorylated on Tyr residues (By similarity). Probably involved in regulation of insulin secretion in pancreatic beta cells (By similarity).
