PYGO1 also known as Pygopus homolog 1 functions as a co-activator in Wnt signaling playing an important role in transcriptional activation. It comprises a protein mass of approximately 44 kDa. The expression of PYGO1 occurs in various human tissues with increased levels observed in the brain and testes. As a component of the β-catenin destruction complex PYGO1 influences the stabilization and nuclear localization of β-catenin which is essential for its activity.

PYGOPUS 1 acts as a significant mediator in the Wnt/β-catenin signaling pathway primarily facilitating gene transcription by interacting with both β-catenin and other transcriptional co-activators.
