Supplementary information

QRICH1 also known as Glutamine-rich protein 1 is a protein that plays an essential role in transcriptional regulation. It has a molecular mass of approximately 91 kDa. This protein is highly expressed in human tissues such as the heart liver and brain. It functions as a transcription factor influencing the expression of genes by binding to specific DNA regions. QRICH1 contains a glutamine-rich region which is typical for transcription factors that assist in the regulation of gene expression.
Biological function summary

QRICH1 modulates cellular processes that involve gene expression. It is part of the transcriptional machinery complex interacting directly with other transcriptional regulators and co-activators. This interaction influences cellular activities such as proliferation differentiation and stress responses. QRICH1 regulates genes involved in cell cycle control and apoptosis affecting how cells respond to various internal and external signals. Its biological role makes it an important target for understanding gene regulatory networks.

Pathways

QRICH1 plays a role in pathways significant to cellular stress responses and apoptosis. It interacts with proteins such as p53 and E2F1 which are important in these pathways. QRICH1 helps mediate cellular responses to DNA damage working alongside p53 in the apoptotic signaling pathway. Additionally its interaction with transcription factors such as E2F1 highlights its involvement in cell cycle regulation and the balance between cell survival and apoptosis.

QRICH1 has connections to cancer and neurodegenerative diseases. In cancer alterations in QRICH1 expression or function can lead to uncontrolled cell proliferation due to its role in regulating genes tied to the cell cycle and apoptosis. Its interaction with p53 and E2F1 further points to its involvement in tumorigenesis when dysregulated. In neurodegenerative disorders QRICH1's expression in the brain tissue suggests it might play a role possibly connected to stress response pathways related to neurodegeneration. Understanding its function could uncover targets or markers for therapeutic interventions in these diseases.

Target data

Transcriptional regulator that acts as a mediator of the integrated stress response (ISR) through transcriptional control of protein homeostasis under conditions of ER stress (PubMed : 33384352). Controls the outcome of the unfolded protein response (UPR) which is an ER-stress response pathway (PubMed : 33384352). ER stress induces QRICH1 translation by a ribosome translation re-initiation mechanism in response to EIF2S1/eIF-2-alpha phosphorylation, and stress-induced QRICH1 regulates a transcriptional program associated with protein translation, protein secretion-mediated proteotoxicity and cell death during the terminal UPR (PubMed : 33384352). May cooperate with ATF4 transcription factor signaling to regulate ER homeostasis which is critical for cell viability (PubMed : 33384352). Up-regulates CASP3/caspase-3 activity in epithelial cells under ER stress. Central regulator of proteotoxicity associated with ER stress-mediated inflammatory diseases in the intestines and liver (PubMed : 33384352). Involved in chondrocyte hypertrophy, a process required for normal longitudinal bone growth (PubMed : 30281152).
