Rab18 also known as RAB18A or RAB18 is a member of the Rab GTPase family a group of proteins involved in intracellular vesicle trafficking. Rab18 is approximately 23 kDa in mass. Researchers find Rab18 expressed widely in various tissues including the brain liver and adipose tissue. As a small GTPase Rab18 switches between an inactive GDP-bound state and an active GTP-bound state regulating membrane trafficking and fusion processes.
Biological function summary

The functions of Rab18 extend beyond simple vesicular transport. Rab18 interacts with lipid droplets and the endoplasmic reticulum influencing lipid metabolism and storage. It is sometimes present in a complex with proteins like TBC1D20 which acts as a GAP (GTPase-activating protein) that further regulates Rab18’s activity. Through these interactions Rab18 influences lipid homeostasis and endoplasmic reticulum organization adding a layer of control to cellular lipid balance.

Pathways

Rab18 participates in the regulation of lipid droplet growth and maturation linking it to lipid metabolism pathways. Its activity plays a role in the tethering of lipid droplets to the endoplasmic reticulum relating it functionally to proteins such as Seipin. The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes (PubMed : 24891604, PubMed : 30970241). Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion (PubMed : 24891604, PubMed : 30970241).
