Supplementary information

RAB1B also known as Ras-related protein Rab-1B is a member of the Rab family of small GTPases. This protein weighs approximately 22.6 kDa and is expressed in the cytoplasm. Mechanically RAB1B regulates vesicular trafficking between the endoplasmic reticulum (ER) and Golgi apparatus. This involves ensuring the proper budding movement and fusion of transport vesicles facilitating efficient transportation of proteins and lipids necessary for cellular function.
Biological function summary

RAB1B plays a role in maintaining cellular homeostasis and protein secretion. It is an important component of the protein transport machinery participating in vesicle coat complex formation. It regulates the movement between the ER and Golgi working closely with other Rab proteins and their effectors to keep the flow of materials steady. The localization and activity of RAB1B influence processes important for cell growth and differentiation.

Pathways

RAB1B operates significantly in the secretory and trafficking pathways. It connects with the Golgi apparatus and ER pathways participating in sorting and cellular signaling. RAB1B interacts with other proteins such as COPII and Golgi SNARE proteins to facilitate transport vesicle movement. These proteins and pathways ensure precise and organized intracellular cargo traffic impacting multiple cellular processes directly tied to the secretory pathway.

Disruptions in RAB1B function can connect to neurodegenerative diseases and cancers. Altered expression can affect protein transport linked to Alzheimer's disease where its dysregulation impacts ER-Golgi pathway proteins like APP. In various cancers abnormal RAB1B levels can influence proliferative signals and cellular stress responses. Understanding RAB1B's role helps to delineate its involvement in these diseases offering targets for therapeutic development.

Target data

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes (PubMed : 20545908, PubMed : 9437002). Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion (PubMed : 9437002). Plays a role in the initial events of the autophagic vacuole development which take place at specialized regions of the endoplasmic reticulum (PubMed : 20545908). Regulates vesicular transport between the endoplasmic reticulum and successive Golgi compartments (By similarity). Required to modulate the compacted morphology of the Golgi (PubMed : 26209634). Promotes the recruitment of lipid phosphatase MTMR6 to the endoplasmic reticulum-Golgi intermediate compartment (By similarity).
