Supplementary information

Rab20 also known as Ras-related protein Rab-20 is a small GTPase belonging to the Rab protein family. Rab20 has a molecular mass of approximately 25 kDa. It is expressed across a variety of tissues with notable presence in the kidney placenta and heart. Rab20's mechanical role involves the regulation of vesicular trafficking operating as a molecular switch by cycling between an active GTP-bound state and an inactive GDP-bound state.
Biological function summary

Rab20 regulates intracellular membrane transport processes. It is involved in endocytic and exocytic transport pathways by controlling vesicle formation movement and fusion. Rab20 participates in the recycling of endosomes contributing to the homeostasis of cellular components. Rab20 is also involved in a complex with other Rab proteins modulating their specific functions in vesicle trafficking.

Pathways

Rab20 plays a role in the endocytic pathway specifically in the recycling endosome pathway where it acts alongside proteins such as Rab11. Rab20 supports the retrieval and recycling of receptors from endosomes back to the plasma membrane. Related to this function Rab20 associates with the exocytic pathway ensuring proper transport of cargo proteins from the Golgi apparatus to the cell surface.

Rab20 has associations with cancer progression and hepatocellular carcinoma. It exhibits altered expression levels in tumors suggesting a role in cell proliferation and metastasis. Furthermore Rab20 may influence autophagic processes in cells impacting conditions such as neurodegenerative disorders. In cancer pathology Rab20 interacts with other proteins such as Ras regulating cell growth and survival pathways.
