The RAB21 protein also known as Ras-related protein Rab-21 plays an important role in vesicle trafficking by regulating endosomal transport. It belongs to the Rab family of small GTPases which are important for vesicle movement and fusion. RAB21 has a molecular mass of approximately 24 kDa. This protein is widely expressed in various tissues including liver kidney and brain indicating its broad role in cellular processes. It localizes to cellular compartments associated with trafficking such as early endosomes and recycling endosomes.

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion (PubMed : 18804435, PubMed : 25648148, PubMed : 31455601). RAB21 is involved in membrane trafficking control (PubMed : 18804435, PubMed : 25648148). During the mitosis of adherent cells, controls the endosomal trafficking of integrins which is required for the successful completion of cytokinesis (PubMed : 18804435). Regulates integrin internalization and recycling, but does not influence the traffic of endosomally translocated receptors in general (By similarity). As a result, may regulate cell adhesion and migration (By similarity). Involved in neurite growth (By similarity). Following SBF2/MTMT13-mediated activation in response to starvation-induced autophagy, binds to and regulates SNARE protein VAMP8 endolysosomal transport required for SNARE-mediated autophagosome-lysosome fusion (PubMed : 25648148).
