Immunocytochemistry/ Immunofluorescence analysis of MCF7 (Human breast adenocarcinoma epithelial cell) cells labeling RAB22A with purified ab137093 at 1/100 dilution (7.6 μg/ml). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) at 1/200 (2.5 μg/ml) dilution. Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1/1000 (2 μg/ml) dilution. DAPI (blue) was used as nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

RAB22A also known as ras-related protein Rab-22A is a small GTPase with a molecular mass of approximately 21 kDa. It belongs to the Rab family of proteins that are involved in vesicular trafficking processes. Expressed in a variety of tissues RAB22A plays an important role in the regulation of endocytic pathways ensuring the proper sorting and trafficking of membrane proteins and lipids. Its expression has been noted prominently in the brain and other tissues indicating its widespread functional relevance.
Biological function summary

RAB22A regulates the early endosomal membrane traffic and acts as an important regulator of intracellular transport. This protein is involved in macropinocytosis and phagocytosis processes essential for cellular nutrient uptake and immune responses. RAB22A participates in protein complexes that modulate membrane fusion and fission impacting cellular dynamics and homeostasis. Through these activities it helps maintain efficient intracellular communication and transport.

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes (PubMed : 16537905, PubMed : 21849477). Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion (PubMed : 16537905). RAB22A plays a role in endocytosis and intracellular protein transport. Mediates trafficking of transferrin/TF from early endosomes to recycling endosomes (PubMed : 16537905).
