RAW 264.7 (mouse macrophage cell line transformed with Abelson murine leukemia virus) cells stained for RAB31 using ab230881 at 1/100 dilution in ICC/IF.

Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight® 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

RAB31 also known as RAB22B belongs to the RAB family which plays an important role in vesicular trafficking. It is a small GTPase with a mass of approximately 23 kDa. RAB31 is mainly expressed in epithelial tissues and certain cancer cell lines. It regulates vesicle formation movement and fusion by cycling between an active GTP-bound state and an inactive GDP-bound state. This cycling is important for its function in the intracellular transport system.

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes (PubMed : 11784320). Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. Required for the integrity and for normal function of the Golgi apparatus and the trans-Golgi network. Plays a role in insulin-stimulated translocation of GLUT4 to the cell membrane. Plays a role in M6PR transport from the trans-Golgi network to endosomes. Plays a role in the internalization of EGFR from the cell membrane into endosomes.
