RAB38 also known as Ras-related protein Rab-38 is a small GTPase involved in vesicular trafficking. It belongs to the RAB family containing proteins important for regulating intracellular vesicle transport. RAB38 has a molecular mass of approximately 25 kDa. This protein is predominantly expressed in lung and melanocytes suggesting its specific roles in these tissues. Its localization plays an important role in the transport and maturation of melanosomes which are organelles responsible for pigment production.
Biological function summary

RAB38 plays a significant role in melanosome biogenesis and function. It acts in concert with other proteins in a larger complex that includes melanophilin and other RAB family members such as RAB27A. This complex influences the transport of melanosomes along microtubules and their positioning within cells. The efficient functioning of RAB38 ensures the proper distribution and secretion of melanin which in turn affects pigmentation in tissues such as skin hair and eyes.

Pathways

RAB38 is involved in two key biological pathways: the lysosome biogenesis pathway and the endocytic recycling pathway. It works alongside proteins such as RAB7 and VPS11 in lysosome-related organelles which regulate endosomal trafficking processes. RAB38's function in these pathways is important for maintaining cellular homeostasis and ensuring the proper degradation and recycling of cellular components. Its role in these pathways demonstrates its interaction with other RAB proteins in modulating vesicle dynamics.

RAB38 has strong associations with Hermansky-Pudlak syndrome type 2 and pulmonary fibrosis. Hermansky-Pudlak syndrome type 2 is characterized by defects in lysosome-related organelles like melanosomes and platelet dense granules. In this disorder RAB38 interacts with proteins like AP3B1 playing a role in the abnormal biogenesis of lysosome-related organelles. In pulmonary fibrosis RAB38's dysregulation may contribute to fibrotic processes in the lung where its expression is notably high affecting processes like inflammation and tissue remodeling. These examples illustrate the involvement of RAB38 in various pathophysiological contexts.
