Rab4/Rab5 effector protein acting in early endocytic membrane fusion and membrane trafficking of recycling endosomes. Required for endosome fusion either homotypically or with clathrin coated vesicles. Plays a role in the lysosomal trafficking of CTSD/cathepsin D from the Golgi to lysosomes. Also promotes the recycling of transferrin directly from early endosomes to the plasma membrane. Binds phospholipid vesicles containing phosphatidylinositol 3-phosphate (PtdInsP3) (PubMed:11062261, PubMed:11788822, PubMed:15020713). Plays a role in the recycling of transferrin receptor to the plasma membrane (PubMed:22308388).

Activity summary

Rabenosyn-5 also known as RBSN or ZFYVE20 is a protein involved in endosomal trafficking. It has a mass of approximately 83 kDa. This protein functions mechanically by binding to early endosomes through its FYVE domain interacting with the small GTPase Rab5. Rabenosyn-5 is expressed in various tissues with higher levels in cells involved in active endocytosis.

Biological function summary

Rabenosyn-5 plays a role in endosomal membrane fusion in part by acting as a tethering effector for early endosomes. It is part of a larger protein complex that includes EEA1 which modulates membrane tethering and fusion events essential for endocytosis and receptor recycling processes. The protein helps facilitate sorting and trafficking of internalized molecules within cells influencing signal transduction and cellular responses to environmental changes.
