RABEP2 also known as Rabaptin-5-like protein or Rabaptin-5b is a protein involved in endosomal trafficking. It has a molecular weight of approximately 50 kDa. RABEP2 functions as an effector protein for Rab GTPases which are critical regulators of membrane traffic in the cell. This protein is widely expressed in various tissues playing key roles in intracellular transport processes. RABEP2 associates with particular endosomal membranes facilitating vesicle budding movement and fusion.
Biological function summary

RABEP2 facilitates membrane trafficking by working in the context of endosomal membrane fusion. It forms part of a larger protein complex that interacts with Rab GTPases especially Rab5 which is a well-known regulator of early endosome dynamics. By binding these GTPases RABEP2 stabilizes active Rab GTPase configurations assisting in their recruitment to endosomal membranes. This mechanism is essential for efficient transport and sorting of internalized molecules.

Pathways

RABEP2 plays a role in cellular pathways involving receptor-mediated endocytosis and vesicle trafficking. In these pathways it connects with the early endosome and sorting endosome pathways which are vital for recycling and degradation processes. RABEP2 works closely with other Rab family proteins like Rab4 and Rab11 sharing roles in ensuring proper receptor allocation and signal transduction. This cooperation enhances the cell's ability to adaptively respond to changes in the extracellular environment.

Modifications or dysfunctions in RABEP2 have associations with cancer and neurodegenerative diseases. Disruptions in Rab GTPase networks including alterations involving RABEP2 can lead to dysfunctional endosomal trafficking which contributes to tumor progression by affecting growth factor receptor recycling. Relationships exist between RABEP2 functionality and other proteins like Rab5 where altered interactions may result in neurodegenerative conditions due to the cumulative effects on synaptic function and neuronal survival.
