RAD51 associated protein 1 also called RAD51AP1 is a protein with a molecular mass of approximately 41 kDa. It interacts directly with RAD51 to assist in DNA repair processes. RAD51AP1 is present in a variety of tissues with higher expression levels seen in testes and ovaries. This protein localizes in the nucleus where it takes part in processes involving DNA repair and stability.
Biological function summary

RAD51AP1 plays an important role in homologous recombination a critical process for the repair of double-strand breaks in DNA. It forms a complex with RAD51 to facilitate the homologous pairing and strand exchange steps of recombination. This interaction helps stabilize the RAD51 nucleoprotein filament which is important for successful recombination events. Apart from RAD51 it also interacts with other proteins involved in DNA repair pathways highlighting its collaborative function in maintaining genomic integrity.

Pathways

RAD51AP1 operates within the DNA damage repair pathway specifically in the homologous recombination repair mechanism. This pathway ensures that DNA double-strand breaks are accurately repaired preventing genomic aberrations. In this pathway the protein works in coordination with RAD51 and other recombinase proteins to facilitate error-free repair. RAD51AP1 also displays a connection to BRCA1 pathways; BRCA1 is another critical player in DNA repair and maintaining genomic stability.

RAD51AP1 has implications in cancer particularly breast and ovarian cancers. Mutations or dysregulation of RAD51AP1 may compromise the DNA repair mechanism leading to increased genomic instability and cancer susceptibility. Furthermore it relates to BRCA1 since both are involved in DNA repair pathways and mutations in BRCA1 are known to significantly elevate cancer risks. Structure-specific DNA-binding protein involved in DNA repair by promoting RAD51-mediated homologous recombination (PubMed : 17996710, PubMed : 17996711, PubMed : 20871616, PubMed : 25288561, PubMed : 26323318). Acts by stimulating D-Loop formation by RAD51 : specifically enhances joint molecule formation through its structure-specific DNA interaction and its interaction with RAD51 (PubMed : 17996710, PubMed : 17996711).
