Immunocytochemistry analysis of HeLa cells labeling RanB1 with ab2937 at 2 μg/mL. Cells were fixed in 4% Paraformaldehyde, permeabilized with 0.1% Triton™ X-100 for 10 minutes and blocked with 1% BSA for 1 hour at RT. Cells were incubated for 3 hours at RT and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate at 1 : 2000 dilution for 45 minutes at RT (Panel A : green). Nuclei (Panel B : blue) were stained with DAPI. F-actin (Panel C : red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin at 1 : 300 dilution. Panel D is a merged image showing cytoplasmic localization. Panel E shows no primary antibody control. The images were captured at 60X magnification.

RanBP1 also known as Ran-binding protein 1 is an important player in the regulation of the Ran GTPase cycle integral to transport processes across the nuclear envelope. This protein has a mass of about 23 kDa. In humans RanBP1 shows expression in various tissues with higher levels observed in testis and brain. It directly interacts with Ran a small GTPase helping to modulate its activity by promoting the conversion between the active GTP-bound form and the inactive GDP-bound form.

Plays a role in RAN-dependent nucleocytoplasmic transport. Alleviates the TNPO1-dependent inhibition of RAN GTPase activity and mediates the dissociation of RAN from proteins involved in transport into the nucleus (By similarity). Induces a conformation change in the complex formed by XPO1 and RAN that triggers the release of the nuclear export signal of cargo proteins (PubMed : 20485264).
