The Ras guanyl-releasing protein 1 (Rasgrp1) also known as CalDAG-GEFII or GRP1 acts as a guanine nucleotide exchange factor (GEF) for Ras and Rap1. These are small GTPases important to intracellular signaling. Rasgrp1 helps convert inactive GDP-bound Ras to its active GTP-bound form which triggers downstream signaling pathways. The protein has a molecular mass of approximately 93 kDa. Rasgrp1 is expressed in various tissues notably in the immune system compartments such as thymus and spleen.
Biological function summary

Rasgrp1 plays an important role in the activation and differentiation of immune cells. It participates in signaling pathways that regulate the function of T-cells and B-cells being critical in their development and response to antigens. Its role is not solitary but part of a larger signaling complex that coordinates the signaling processes initiated by the T-cell receptor. Such activities enable proper immune responses and control the immune system balance.

Pathways

Rasgrp1 integrates into the Ras-MAPK and Rap1 signaling pathways which have pivotal roles in cell growth differentiation and survival. Within these pathways it interacts with related proteins such as Sos (Son of Sevenless) and Raf kinases. The involvement of Rasgrp1 in the Ras-MAPK pathway exemplifies its importance in modulating cellular responses to external stimuli especially those related to immune system activation.

Rasgrp1 mutations or dysregulation associate with immune system disorders like systemic lupus erythematosus (SLE) and some forms of immunodeficiency. Alterations in Rasgrp1 expression can disrupt normal immune function leading to autoimmune conditions. Connections to these diseases often highlight interactions with proteins like Bcl10 and CARMA1 which also participate in the immune response regulation and signal transduction associated with these disorders.
