Rb2 p130 also known as Retinoblastoma-like protein 2 functions as an important regulator of cell cycle progression. It has a molecular weight of about 130 kDa and members of the retinoblastoma (Rb) protein family which also includes p107 and RB1. These proteins help control the transition from the G1 to S phase during the cell cycle. Rb2 p130 is widely expressed in various tissues particularly in quiescent cells where it plays a role in maintaining cells in the G0 phase.
Biological function summary

The protein acts as a fundamental component of the DREAM (DP RB-like E2F and MuvB) complex which plays an important role in repressing the transcription of E2F target genes. This repression ensures the proper regulation of cell proliferation and differentiation. The protein contributes to maintaining genomic stability by helping to control the entry of cells into the DNA synthesis phase thereby acting as a tumor suppressor.

Pathways

Rb2 p130 is heavily involved in the cell cycle control and proliferation pathways. It associates with the E2F family of transcription factors which are critical in transcription regulation of genes important for cell cycle progression. It modulates activities within the p53 pathway as well another pathway important for cell cycle arrest and apoptosis. This relation highlights its interplay with proteins like cyclin-dependent kinases (CDKs) which regulate cell cycle transitions through phosphorylation events.

The protein has been linked to several cancer types due to its role as a tumor suppressor. Disruptions in its expression or function can lead to unchecked cellular division paving the way for tumorigenesis. Mutations or alterations in the Rb pathway involving Rb2 p130 RB1 and p107 can contribute to the development of cancers such as retinoblastoma and lung carcinoma. The involvement of these proteins signifies their importance in maintaining controlled cell division with impairments correlating strongly to cancer progression.
