Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human lung cancer with alkaline phosphatase treatment (image A) and treated without alkaline phosphatase (image B) tissue sections labeling Rb2 p130 with purified ab68136 at 1/600 dilution (0.36 μg/ml). Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0). ImmunoHistoProbe one step HRP Polymer (ready to use) was used as the secondary antibody. Biological function summary
The protein acts as a fundamental component of the DREAM (DP RB-like E2F and MuvB) complex which plays an important role in repressing the transcription of E2F target genes. This repression ensures the proper regulation of cell proliferation and differentiation. The protein contributes to maintaining genomic stability by helping to control the entry of cells into the DNA synthesis phase thereby acting as a tumor suppressor.
Pathways
Rb2 p130 is heavily involved in the cell cycle control and proliferation pathways. It associates with the E2F family of transcription factors which are critical in transcription regulation of genes important for cell cycle progression. It modulates activities within the p53 pathway as well another pathway important for cell cycle arrest and apoptosis. This relation highlights its interplay with proteins like cyclin-dependent kinases (CDKs) which regulate cell cycle transitions through phosphorylation events.
