Rbm15 also known as OTT and RBM15B are members of the RNA-binding motif protein family. Rbm15 has a molecular mass of approximately 101 kDa. Both proteins possess RNA-binding motifs that mediate their involvement in post-transcriptional regulation. Rbm15 and RBM15B are widely expressed in various tissues with notable expression in hematopoietic cells. They regulate RNA methylation by associating with the m6A methyltransferase complex influencing mRNA processing and nuclear export.
Biological function summary

Rbm15 and RBM15B play significant roles in hematopoiesis and embryonic development. They are known to be components of the m6A methylation complex which modifies adenosine residues on RNA affecting RNA stability and translation. This complex interaction enables them to participate in controlling cell differentiation and growth important in maintaining proper cellular function and development. Through their RNA binding ability they also influence splice site selection and alternative splicing of pre-mRNA which is essential in the generation of protein diversity.

Pathways

Both Rbm15 and RBM15B contribute significantly to the maintenance of the Notch signaling and Wnt/β-catenin signaling pathways. These pathways are central to cell fate determination proliferation and apoptosis. Rbm15 interacts with key proteins like Notch receptors promoting signal mediation and thereby affecting downstream gene expression. In the Wnt pathway RBM15B may interface with β-catenin to regulate transcriptional activity influencing cell cycle and differentiation processes alongside proteins such as LEF/TCF.

Rbm15 and RBM15B have connections with acute myeloid leukemia (AML) and developmental disorders. Rbm15 is known for its implication in AML through fusion with the MKL1 gene contributing to leukemogenesis by altering gene expression profiles. Abnormal m6A methylation patterns involving RBM15B can disrupt normal cell differentiation and have been linked to developmental anomalies.
