RNA-binding protein involved in the regulation of smooth muscle cell differentiation and proliferation in the gastrointestinal system (PubMed:25064856). Binds NOG mRNA, the major inhibitor of the bone morphogenetic protein (BMP) pathway. Mediates an increase of NOG mRNA levels, thereby contributing to the negative regulation of BMP signaling pathway and promoting reversible dedifferentiation and proliferation of smooth muscle cells (By similarity). Acts as a pre-mRNA alternative splicing regulator (By similarity). Mediates ACTN1 and FLNB alternative splicing (By similarity). Likely binds to mRNA tandem CAC trinucleotide or CA dinucleotide motifs (PubMed:24860013).

The RNA-binding protein with multiple splicing 2 commonly known as RBPMS2 or HERMES plays an important mechanical role as a regulator of RNA processing. With a molecular mass of approximately 25 kDa RBPMS2 acts in various tissues including heart eye and smooth muscle cells. Its expression is often specific and changes dynamically during development and in response to different cellular signals hinting at a control function in tissue-specific gene expression.
By binding to RNA RBPMS2 influences mRNA stability and splicing directly affecting gene expression levels in cells. RBPMS2 is known to be involved in the formation of complexes with other proteins that regulate RNA splicing and transport. Its involvement in these complexes emphasizes its contribution to maintaining proper cellular function and development which is critical in the context of muscle tissue and eye development.
RBPMS2 participates in essential signaling pathways such as Wnt signaling and TGF-beta signaling where it interacts with components vital for transcriptional regulation and cytoskeletal organization. This protein's interactions with factors like SMADs in the TGF-beta pathway and beta-catenin in the Wnt pathway help modulate key events in cell proliferation and differentiation illustrating its integrative role in cellular signaling networks.
Studies suggest that RBPMS2 is linked to conditions such as cataracts and cardiomyopathies. Mutations or dysregulation in RBPMS2 expression can disrupt normal cellular activities contributing to disease progression. Protein interactions such as those with SMAD family are significant in understanding its role in abnormal tissue development and associated pathologies offering potential insights into therapeutic intervention strategies.
