REEP5 also known as Receptor Expression Enhancing Protein 5 facilitates cell machinery in organizing and shaping the membranes of the endoplasmic reticulum. This protein has a molecular mass of approximately 22 kDa. REEP5 is expressed abundantly in the heart and skeletal muscle though it also exists in other tissues at lower levels. The protein is known to assist in membrane topology which is essential for maintaining proper endoplasmic reticulum morphology and function.
Biological function summary

REEP5 supports endoplasmic reticulum tubule formation helping maintain the high curvature structure of the organelle. It becomes part of a larger protein complex contributing to the development and maintenance of the fine network structure typical of the endoplasmic reticulum. By doing so REEP5 plays a role in the spatial organization of the cellular environment ensuring the proper segregation and function of cellular components.

Pathways

REEP5 actively participates in cellular stress response and the unfolded protein response pathway. This pathway involves proteins such as IRE1 and PERK which sensor and manage cellular stress by regulating protein folding and assisting in maintaining homeostasis. REEP5 provides structural regulation in these pathways impacting protein synthesis and secretion processes which are essential for adapting to cellular demands and stress.

REEP5 has been implicated in heart conditions particularly dilated cardiomyopathy. This disorder involves the heart muscle becoming weakened and enlarged affecting its ability to pump blood efficiently. Mutations or dysregulation of REEP5 impact its interaction with proteins like ATL1 and ATL2 which are involved in membrane fusion processes. Additionally REEP5 has shown associations with neurodegenerative conditions indicating its wider significance in maintaining cellular health and function.
