REP-2 also known by some researchers as Rab escort protein 2 acts mechanically by facilitating the proper prenylation and trafficking of Rab proteins. These Rab proteins are small GTPases essential for vesicular transport processes. REP-2 has a molecular weight of approximately 75 kDa and is expressed in various tissues with particularly high levels in the brain and pancreas. Its main role involves binding to and transferring newly synthesized Rab proteins to the Rab geranylgeranyltransferase complex for prenylation.
REP-2 functions in processes involving Rab protein maturation and membrane trafficking. It is a critical component of the Rab escort complex working alongside other members to stabilize the formation of Rab-GTP which subsequently influences intracellular vesicle transport. By ensuring the proper localization and function of Rab GTPases REP-2 supports numerous cellular processes including endocytosis exocytosis and organelle motility which are fundamental to regular cell operation.
REP-2 operates within the vesicular trafficking pathways that are vital for cellular homeostasis and signal transduction. It forms part of the Rab protein cycle and interacts closely with the Rab geranylgeranyltransferase helping the prenylation of newly made Rab proteins. This function ties it to the broader network of proteins including Rab1 and Rab3 where REP-2 assists in securing their role in the intricate machinery of vesicular movement and fusion within the cell.
REP-2 has connections to certain neurological and metabolic diseases. It has been linked with choroideremia a retinal degenerative disease where dysfunctional REP-2 leads to impaired Rab function. Such dysfunction results in defective vesicular transport causing the gradual degeneration of photoreceptors. Additionally mutations affecting REP-2 relate to some neurodegenerative disorders highlighting its interaction with Rab27a another protein integral to similar cellular pathways.
