Flow Cytometry (Intracellular) - Anti-Resistin antibody [EPR23334-171] (AB275878)
Intracellular flow cytometric analysis of 4% paraformaldehyde-fixed, 90% methanol-permeabilized U937 (human histiocytic lymphoma monocyte) cells labelling Resistin with ab275878 at 1/500 dilution (0.1μg)/ (Red) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Black) and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue).
A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/2000 dilution was used as the secondary antibody.

Immunoprecipitation - Anti-Resistin antibody [EPR23334-171] (AB275878)
Resistin was immunoprecipitated from 0.35 mg U937 (human histiocytic lymphoma monocyte) whole cell lysate with ab275878 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab275878 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/1000 dilution.
Lane 1 : U937 whole cell lysate 10 μg
Lane 2 : ab275878 IP in U937 whole cell lysate
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab275878 in U937 whole cell lysate
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 5.5 seconds.

Biological function summary
Resistin affects glucose homeostasis and has been shown to modulate insulin resistance. It does not function as part of a larger protein complex but instead acts through specific receptors on cell surfaces. Resistin influences several cellular processes including energy balance and the modulation of inflammatory responses.
Pathways
Several processes contribute to the detailed function of resistin. It is involved in the insulin signaling pathway and the inflammatory pathway. Resistin interacts with proteins and receptors like TLR4 which have key roles in inflammatory response signaling and metabolic regulation. These interactions can disrupt insulin signaling and glucose uptake in many cell types.
