Retinal protein 4 also known as RBP4 is a transportation protein that plays an important role in transporting vitamin A also known as retinol from the liver to peripheral tissues. RBP4 has a molecular mass of approximately 21 kDa. It is mostly expressed in the liver and secreted into the bloodstream. Besides the liver RBP4 can be found in other tissues such as the adipose tissue and kidneys. It forms complexes with transthyretin that prevent the renal filtration of the retinol-RBP4 complex thereby allowing effective retinol transportation.
Biological function summary

Retinal-binding protein 4 facilitates the delivery of vitamin A to cells which is essential for vision and cellular differentiation. This protein forms a complex with retinol binding it tightly to deliver to target tissues. Vitamin A is important for the synthesis of the visual pigment in photoreceptor cells and influences gene expression particularly in epithelial cells which underlines its involvement in the visual and cellular maintenance functions.

Pathways

Retinal protein 4 is integral to the visual cycle and vitamin A metabolism pathways. It interacts with other proteins such as STRA6 a receptor on the cell surface that allows vitamin A uptake when RBP4 binds to it. Moreover its role in the vitamin A signaling pathway showcases its necessity for maintaining optimal physiological levels of retinoic acid vital in various developmental processes.

Retinal protein 4 has associations with type 2 diabetes and retinitis pigmentosa. Elevated levels of RBP4 have been linked to insulin resistance contributing to metabolic syndrome components like type 2 diabetes. Additionally mutations affecting RBP4 can lead to an impaired visual cycle manifesting as retinitis pigmentosa a hereditary disorder causing progressive vision loss. Its interaction with proteins like retinoid isomerohydrolase (RPE65) points to its significance in the visual impairment seen in such disorders.

Involved in synaptic functions in photoreceptor cells, the signal transduction in immune cells as a Src family kinase activator, endosome recycling, the uptake of bacteria and endocytosis, protein trafficking in sensory neurons and as lipid-binding chaperone with specificity for a diverse subset of myristoylated proteins. Specifically binds the myristoyl moiety of a subset of N-terminally myristoylated proteins and is required for their localization. Binds myristoylated GNAT1 and is required for G-protein localization and trafficking in sensory neurons. Probably plays a role in trafficking proteins in photoreceptor cells. Plays important roles in mediating Src family kinase signals for the completion of cytokinesis via RAB11A.
