Supplementary information

The retinoblastoma-binding protein 6 (RBBP6) also known as P2 is a multifunctional protein with a molecular mass of approximately 200 kDa. RBBP6 interacts with various proteins such as p53 and the retinoblastoma protein (Rb). It participates in processes like transcription splicing and ubiquitination. Cells express RBBP6 in tissues across multiple organs with significant levels present in the liver heart and lung tissues. The expression pattern indicates a wide role in cellular functions.
Biological function summary

RBBP6 functions as part of a complex involved in cell cycle regulation and apoptosis. It interacts with the p53 tumor suppressor protein facilitating its regulation and influences various cellular processes. Additionally RBBP6 is implicated in RNA processing due to its association with the 3'-end processing of mRNA. Through these roles RBBP6 ensures proper cellular response to stress and DNA damage connecting it to mechanisms of maintaining genomic stability.

Pathways

RBBP6 plays a significant role in the p53 signaling and ubiquitin-proteasome pathways. Within the p53 pathway RBBP6 modulates p53 activity by affecting its degradation; this interaction impacts cell cycle arrest and apoptosis. It also contributes to the ubiquitin-proteasome system by labeling proteins for degradation which maintains protein quality and turnover. These pathways show RBBP6's pivotal function in controlling cell proliferation and survival.

Anomalies in RBBP6 expression or function link to cancer and retinoblastoma. Mutations in RBBP6 can disrupt its interaction with p53 and Rb proteins leading to uncontrolled cell proliferation and tumorigenesis. The protein's dysfunction may also contribute to the development of retinoblastoma a pediatric eye cancer as improper Rb protein regulation compromises cell cycle control. Understanding RBBP6’s role in these diseases offers potential for targeted therapies in oncology.

Target data

E3 ubiquitin-protein ligase which promotes ubiquitination of YBX1, leading to its degradation by the proteasome (PubMed : 18851979). May play a role as a scaffold protein to promote the assembly of the p53/TP53-MDM2 complex, resulting in increase of MDM2-mediated ubiquitination and degradation of p53/TP53; may function as negative regulator of p53/TP53, leading to both apoptosis and cell growth (By similarity). Regulates DNA-replication and the stability of chromosomal common fragile sites (CFSs) in a ZBTB38- and MCM10-dependent manner. Controls ZBTB38 protein stability and abundance via ubiquitination and proteasomal degradation, and ZBTB38 in turn negatively regulates the expression of MCM10 which plays an important role in DNA-replication (PubMed : 24726359).. (Microbial infection) [Isoform 1] : Restricts ebolavirus replication probably by impairing the vp30-NP interaction, and thus viral transcription.
