Supplementary information

Retinoic Acid Receptor gamma 1 (RARG) functions as a nuclear receptor and transcription factor which means it binds to specific DNA sequences to regulate gene expression. Also known as RAR gamma this protein has a molecular mass of approximately 51 kDa. RARG is expressed in various tissues most notably in skin lung and the central nervous system. It acts by binding to retinoic acid a derivative of vitamin A which is essential for modulating cell differentiation and growth.
Biological function summary

Retinoic Acid Receptor gamma 1 influences the regulation of several developmental processes and cellular homeostasis. The receptor operates often as part of a heterodimeric complex with the Retinoid X Receptor (RXR). This interaction is important for the receptor's ability to bind to retinoic acid response elements in DNA driving the expression of genes that control cell proliferation apoptosis and development. Its activity impacts skin keratinization and neural differentiation significantly.

Pathways

Retinoic Acid Receptor gamma 1 is key in the retinoic acid signaling pathway which orchestrates the transcription of essential genes linked to cellular differentiation and organogenesis. RARG is closely related to other retinoic acid receptors like RAR alpha and RAR beta in this pathway. These receptors facilitate a network where retinoic acid acts as a ligand initiating changes in gene expression that affect cellular response and fate.

The dysregulation of RARG links to health issues such as skin disorders and certain types of cancer. Psoriasis for example involves abnormal proliferation and differentiation of keratinocytes where RARG may play a role alongside other proteins like RXR. In leukemia alterations in RARG have associations with disrupted differentiation of blood cell lines closely interacting with oncogenic fusion proteins. Understanding RARG's involvement in these conditions helps in developing targeted treatments.

Target data

Receptor for retinoic acid. Retinoic acid receptors bind as heterodimers to their target response elements in response to their ligands, all-trans or 9-cis retinoic acid, and regulate gene expression in various biological processes. The RAR/RXR heterodimers bind to the retinoic acid response elements (RARE) composed of tandem 5'-AGGTCA-3' sites known as DR1-DR5. In the absence of ligand, acts mainly as an activator of gene expression due to weak binding to corepressors. Required for limb bud development. In concert with RARA or RARB, required for skeletal growth, matrix homeostasis and growth plate function (By similarity).
