Retinol dehydrogenase 16 (RDH16) also known as PED1 plays an important role in the oxidation of retinol to retinal. This enzyme has a mass of approximately 35 kDa. RDH16 is expressed mainly in the liver as well as in the kidney and intestine where it facilitates the metabolism of retinoids essential compounds for various biological processes. RDH16 acts by converting all-trans-retinol to all-trans-retinal a critical step in retinoid metabolism.
Biological function summary

RDH16 contributes significantly to the biosynthesis of retinoic acid a derivative of vitamin An important for cell differentiation and proliferation. While RDH16 does not form part of a larger known complex its activity directly influences the levels of retinoic acid affecting signaling pathways involved in embryonic development vision and immune function. These interactions highlight RDH16's role in modulating important cellular functions through retinoid metabolism.

Pathways

The enzymatic activity of RDH16 integrates into the retinoic acid signaling and metabolic pathways. This activity involves interconversions in the retinoid cycle that are necessary for the availability of retinoic acid in tissues. Oxidizes all-trans-retinol, 9-cis-retinol, 11-cis-retinol and 13-cis-retinol to the corresponding aldehydes (PubMed : 10329026, PubMed : 12534290, PubMed : 9677409). Has higher activity towards CRBP-bound retinol than with free retinol (PubMed : 12534290). Oxidizes also 3-alpha-hydroxysteroids.
