REV3L also known as DNA polymerase zeta catalytic subunit or hREV3 is an essential component of the DNA polymerase zeta (Pol ζ) complex. This protein has an approximate molecular weight of 353 kDa. It carries out its functions in several tissues with main expression in the testis thymus and ovaries although it appears in other tissues as well. Mechanically REV3L serves as the catalytic component in DNA synthesis particularly facilitating translesion DNA synthesis which allows DNA replication to bypass lesions or damage.
Biological function summary

DNA damage tolerance mechanisms involve REV3L where it plays a critical role as part of the Pol ζ complex. Pol ζ operates by extending DNA synthesis across non-instructive damages that disrupt the DNA double helix. This action involves error-prone processes but remains necessary for maintaining genomic integrity. REV3L binds with the REV7 protein in this complex to perform these translesion DNA synthesis actions shielding cells from potentially lethal damage or genomic instability due to unrepairable DNA lesions.
