Western blot - Anti-Rex1 antibody [RP23040041] (AB308003)
Western blot analysis of Rex1 was performed by loading 20 µg of HeLa, 3T3, NRK, MDCK, and Cos7 whole cell lysates and 5uL of prestained protein ladder per well onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% milk in TBST for 1 hour at room temperature. Rex1 was detected at ~35 kD using ab308003 at a concentration of 2 µg/mL in 5% milk in TBST overnight at 4C on a rocking platform, followed by a Goat anti-Rabbit IgG-HRP secondary antibody at a concentration of 1 µg/mL for at least 30 minutes at room temperature. Chemiluminescent detection was performed.
All lanes:
Western blot - Anti-Rex1 antibody [RP23040041] (ab308003) at 2 µg/mL
Lane 1:
HeLa whole cell lysate at 20 µg
Lane 2:

What are recombinant multiclonals?
Recombinant multiclonals are a mixture of recombinant antibodies co-expressed from a library of heavy and light chains. They offer several advantages including:
- - The sensitivity of polyclonal antibodies by recognising multiple epitopes
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free batch production

Biological function summary
Rex1 influences stem cell pluripotency and differentiation. It is part of the network of pluripotency factors along with Oct4 Sox2 and Nanog. These proteins form a regulatory circuit that maintains embryonic stem cell identity. Rex1 directly interacts with chromatin structure facilitating the accessibility of specific genes related to stem cell maintenance and identity. The expression levels of Rex1 have a significant impact on the transition between pluripotency and differentiated states within cells.
Pathways
Rex1 is involved in key signaling pathways regulating pluripotency like the LIF/STAT3 and Wnt signaling pathways. Within these pathways Rex1 collaborates with other transcription factors to promote the expression of genes required for stem cell growth and maintenance. Rex1 and Nanog work closely in these pathways to sustain the pluripotent status of stem cells by activating necessary genetic programs and preventing differentiation signals.
