Western blot analysis of Rex1 in whole cell extracts of A431 (lane 1) and rat kidney lysate (lane 2) using ab308003 at a dilution of 1 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~35kDa.

Western blot analysis of Rex1 was performed by loading 20 µg of HeLa, 3T3, NRK, MDCK, and Cos7 whole cell lysates and 5uL of prestained protein ladder per well onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% milk in TBST for 1 hour at room temperature. Rex1 was detected at ~35 kD using ab308003 at a concentration of 2 µg/mL in 5% milk in TBST overnight at 4C on a rocking platform, followed by a Goat anti-Rabbit IgG-HRP secondary antibody at a concentration of 1 µg/mL for at least 30 minutes at room temperature. Chemiluminescent detection was performed.

Rex1 also known as Zfp42 is a transcription factor and zinc finger protein with a mass of approximately 42 kDa. This protein is expressed mostly in embryonic stem cells and some adult stem cells. Rex1 functions by binding to DNA sequences influencing gene expression that is essential for maintaining stem cell self-renewal and pluripotency. As a result Rex1 plays an important role in regulating cellular processes related to developmental potential.
Biological function summary
