The elongation of primed DNA templates by DNA polymerase delta and epsilon requires the action of the accessory proteins PCNA and activator 1. This subunit binds to the primer-template junction. Binds the PO-B transcription element as well as other GA rich DNA sequences. Could play a role in DNA transcription regulation as well as DNA replication and/or repair. Can bind single- or double-stranded DNA. Interacts with C-terminus of PCNA. 5' phosphate residue is required for binding of the N-terminal DNA-binding domain to duplex DNA, suggesting a role in recognition of non-primer template DNA structures during replication and/or repair.

The Reduced Folate Carrier 1 (RFC1) also known as SLC19A1 is a membrane protein that facilitates the transport of reduced folates and antifolate drugs into the cell. It exhibits a mass of approximately 65 kDa. RFC1 functions as an integral part of the cell membrane predominantly expressed in epithelial tissues including the intestine and placenta. It acts as a folate transporter that enables cellular uptake of folates critical for DNA synthesis and repair nucleotide biosynthesis and methylation reactions.
RFC1 plays a major role in maintaining intracellular concentrations of folates a necessity for cell division and growth. It is part of a broader folate transport system ensuring proper utilization of folates in metabolic processes. The protein functions alongside the Proton-Coupled Folate Transporter (PCFT) and the Folate Receptor (FR) which work in tandem yet accommodate different pH levels and folate forms.
RFC1 is essential to the folate metabolic pathway ensuring the movement of 5-methyltetrahydrofolate a critical form of folate across cell membranes. It engages in interactions with proteins like methionine synthase which play a role in homocysteine remethylation to methionine. This pathway is indispensable to methyl group transfer reactions impacting genome stability and gene expression.
Dysfunction in RFC1 activity associates with conditions such as hereditary folate malabsorption and increased susceptibility to certain cancers. Its impaired function can lead to low folate uptake causing severe consequences in rapidly dividing cells. Evidence indicates that alterations in RFC1 activity can influence the effectiveness of antifolate chemotherapeutic agents involving proteins like thymidylate synthase that depend on folate availability.
