Supplementary information

RFRP also known as RFamide-related peptide ranges in mass from 1.38 kDa to 2.85 kDa depending on the specific peptide variant. RFRP is expressed in the central nervous system particularly in the hypothalamus and pituitary gland and also found within the testis and ovary tissues. It originates from the VGF peptide precursor and plays a role in modulating various physiological processes.
Biological function summary

The RFamide-related peptide influences reproductive functions such as the regulation of gonadotropin release. It functions as an inhibitory factor on the secretion of gonadotropin-releasing hormone (GnRH) working in tandem with other neuropeptides and factors. RFRP is not a part of a larger complex but interacts closely with related neurohormones to exert its physiological effects.

Pathways

Several neuroendocrine control systems involve the RFamide-related peptide. RFRP plays an important role in the hypothalamic-pituitary-gonadal (HPG) axis where it modulates the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). It interacts with receptor proteins like GPR147 which influences these essential pathways and subsequently affects reproductive system functionality.

The RFamide-related peptide has links to reproductive health issues such as delayed puberty and infertility. Abnormal activities or levels of RFRP can interfere with hormonal balance making it a potential target for addressing these reproductive system disorders. Additionally its regulation of GnRH suggests possible connections to disorders involving altered hormonal cycles including polycystic ovary syndrome (PCOS) where excessive or insufficient RFRP activity can disrupt cyclical hormone release patterns.

Target data

Neuropeptide RFRP-1. Efficiently inhibits forskolin-induced production of cAMP. Acts as a potent negative regulator of gonadotropin synthesis and secretion (PubMed : 11025660, PubMed : 20027225). Induces secretion of prolactin (By similarity).. Neuropeptide NPVF. Efficiently inhibits forskolin-induced production of cAMP. Blocks morphine-induced analgesia.. Neuropeptide RFRP-2. Shows no inhibitory activity of forskolin-induced production of cAMP.
