RFX3 functions as a transcription factor binding specifically to X-box motifs within DNA influencing the expression of various genes important for cell processes. This protein is present in a variety of tissues including the brain kidney and pancreas suggesting its diverse and widespread role in cellular activities.
Biological function summary

The function of RFX3 involves the modulation of genes critical for cilia development and function. RFX3 forms part of a gene regulatory complex that oversees ciliogenesis. Through this complex it controls the transcription of genes essential for the formation of motile and primary cilia. The activity of RFX3 maintains proper cilia structure and movement essential for cellular signaling and environmental sensing.

Pathways

The involvement of RFX3 focuses largely on the ciliogenesis and hedgehog signaling pathways. RFX3 regulates genes necessary for the assembly and function of cilia linking it to the hedgehog signaling pathway where cilia play an important role in signal reception and transduction. RFX3 interacts with proteins like RFX1 and RFX2 which share similar DNA-binding domains highlighting its preserved function across these pathways.

Disruptions in RFX3 can relate to ciliopathies and diabetes. Ciliopathies such as Joubert syndrome arise from defective ciliogenesis due to compromised RFX3 function with related proteins like RFX1 also being implicated. Diabetes connects to RFX3 through its expression in pancreatic beta cells suggesting a role in insulin regulation and glucose metabolism. Transcription factor required for ciliogenesis and islet cell differentiation during endocrine pancreas development. Essential for the differentiation of nodal monocilia and left-right asymmetry specification during embryogenesis. Required for the biogenesis of motile cilia by governing growth and beating efficiency of motile cells. Also required for ciliated ependymal cell differentiation. Regulates the expression of genes involved in ciliary assembly (DYNC2LI1, FOXJ1 and BBS4) and genes involved in ciliary motility (DNAH11, DNAH9 and DNAH5) (By similarity). Together with RFX6, participates in the differentiation of 4 of the 5 islet cell types during endocrine pancreas development, with the exception of pancreatic PP (polypeptide-producing) cells. Regulates transcription by forming a heterodimer with another RFX protein and binding to the X-box in the promoter of target genes (PubMed : 20148032). Represses transcription of MAP1A in non-neuronal cells but not in neuronal cells (PubMed : 12411430).
