RGL2 also known as Ral Guanine Nucleotide Dissociation Stimulator-Like 2 is a protein that functions importantly in the regulation of small GTPases. It has a mass of approximately 109 kDa. RGL2 is expressed in various tissues but high levels are often present in the nervous tissue and the endocrine system. This expression pattern suggests its involvement in neuronal signaling and hormonal regulation.
Biological function summary

Ral Guanine Nucleotide Dissociation Stimulator-Like 2 acts as a guanine nucleotide exchange factor (GEF) which activates Ral GTPases by facilitating the exchange of GDP for GTP. The protein is a member of the RAS subfamily GEFs and contributes greatly to the formation of signaling complexes that affect multiple cellular processes. RGL2 interacts with other proteins to modulate cell growth cytoskeletal organization and vesicle trafficking.

Pathways

RGL2 plays a role in the MAPK/ERK signaling pathway and the PI3K/Akt pathway. These pathways are critical for cell proliferation and survival. It works closely with RalA and RalB proteins acting as a mediator in transmitting signals from receptor tyrosine kinases. Through this interaction RGL2 contributes to the regulation of cell cycle progression and apoptosis.

RGL2 has associations with cancer and neurodegenerative diseases. In some cancers upregulation of RGL2 leads to uncontrolled cellular proliferation where the protein interacts with other members of the Ras signaling pathway like c-Raf. Furthermore RGL2's role in neurodegenerative diseases is revealed by its impact on neuronal signaling potentially influencing conditions such as Alzheimer's disease through dysfunction in vesicular trafficking and synaptic plasticity.
