Supplementary information

RGL3 also known as Ral guanine nucleotide dissociation stimulator-like 3 is a protein involved in regulating various cellular processes. It has a molecular weight of approximately 85 kDa. RGL3 is expressed in multiple tissues including the brain and liver indicating its involvement in diverse physiological functions. The protein interacts mechanically with other cellular proteins to promote GDP-GTP exchange on Ral GTPases which act as molecular switches in numerous signaling pathways.
Biological function summary

The protein RGL3 plays a role in cellular signaling and transformation. It is a part of the RalGDS protein family and functions within a complex that includes RalA and RalB GTPases. This complex is instrumental in directing cellular events that involve rearrangements in the cytoskeleton and membrane trafficking. RGL3 also impacts gene expression by modulating the activity of its associated GTPases which influence cell growth and differentiation.

Pathways

RGL3 is intricately linked to the Ras signaling pathway and the MAPK/ERK cascade. In the Ras pathway RGL3 associates with Ras proteins influencing cellular proliferation and survival. In the MAPK/ERK cascade RGL3 indirectly affects the downstream phosphorylating actions by modifying the activity of Ral GTPases thereby contributing to the regulation of mitogenic signals. These interactions reflect RGL3's significance in cellular signal transduction and communication.

Disruptions in RGL3 function have implications in certain cancers such as colorectal cancer where altered signaling dynamics lead to uncontrolled cell growth. RGL3's role in the Ras pathway links it to oncogenic transformation where mutations of Ras proteins result in aberrant signaling. Additionally there is evidence suggesting a relation to diabetes through its impact on insulin signaling pathways influencing glucose metabolism. The interplay between RGL3 and Ras proteins along with other molecules in these pathways highlights its potential impact on disease pathogenesis.
