RGP1 also known as the RAB6A GTPase Activating Protein 1 is a protein involved in the activation of Rab GTPases particularly Rab6A. It has a molecular mass of approximately 46 kDa. RGP1 primarily expresses in the Golgi apparatus where it plays a role in membrane trafficking and vesicle transport. It acts as an important component in regulating the functionality of the Golgi complex which is essential for maintaining intracellular trafficking.
Biological function summary

The protein interacts with several other proteins and acts in a complex with RIC1. RGP1 plays an important role in tethering and fusion events during late Golgi transport. Its function modulates the movement of vesicles between the Golgi and endosome impacting cell transport processes significantly. The complex is necessary for maintaining the accuracy of cargo delivery and removal making it important for cellular homeostasis and normal cell growth.

Pathways

RGP1 serves a role in the retrograde transport pathway guiding the efficient movement of proteins from the Golgi back to the endoplasmic reticulum. The RIC1-RGP1 complex acts as a guanine nucleotide exchange factor (GEF), which activates RAB6A by exchanging bound GDP for free GTP and may thereby required for efficient fusion of endosome-derived vesicles with the Golgi compartment. The RIC1-RGP1 complex participates in the recycling of mannose-6-phosphate receptors.
