Regulates G protein-coupled receptor signaling cascades, including signaling via muscarinic acetylcholine receptor CHRM2 and dopamine receptor DRD2. Inhibits signal transduction by increasing the GTPase activity of G protein alpha subunits, thereby driving them into their inactive GDP-bound form (PubMed:15096504). Binds selectively to GNAZ and GNAI2 subunits, accelerates their GTPase activity and regulates their signaling activities. Negatively regulates mu-opioid receptor-mediated activation of the G-proteins (By similarity).

Activity summary

RGS17 also known as RGSZ2 acts as a regulator of G-protein signaling. It belongs to the RGS protein family which plays a critical role by accelerating the GTPase activity of G alpha subunits therefore terminating G-protein-coupled receptor (GPCR) signaling pathways. RGS17 has a molecular mass of approximately 27 kDa and is mainly expressed in the brain specifically in regions such as the cerebellum and nucleus accumbens. It is also found in other tissues like the lung and placenta.

Biological function summary

RGS17 influences various cellular processes through its ability to control GPCR signaling. It functions as part of a signaling complex that acts to quickly switch off G-protein signals therefore ensuring that cells can respond appropriately to new stimuli. This regulation is particularly important in neuronal cells where rapid signaling changes are essential for proper function. In addition RGS17 participates in the modulation of neurotransmitter activities which affects processes like mood and perception.

Pathways

RGS17 is involved in the regulation of both the G-protein signaling pathway and the cAMP signaling pathway. It interacts closely with G alpha subunits such as Gαo Gαi and Gαz which play key parts in these pathways. Associated diseases and disorders

Research suggests a link between RGS17 and certain types of cancer including lung and prostate cancer.
