RGS20 also known as Regulator of G-protein Signaling 20 is a protein that functions mechanically as a GTPase-activating protein (GAP) for G-alpha subunits of heterotrimeric G proteins. It accelerates the termination of G-protein coupled receptor (GPCR) signaling pathways by increasing the GTPase activity of the G-alpha subunit. The protein has a molecular mass of approximately 26 kDa. RGS20 is expressed in various tissues with high levels in the brain and some expression in peripheral tissues such as the spleen and lung.
Biological function summary

RGS20 modulates signal transduction by accelerating the deactivation of G-proteins which plays an important role in regulating GPCR signaling. It associates with G-beta and G-gamma forming a complex that allows for more efficient signal regulation. The activity of RGS20 is vital in neuronal signaling processes within the brain contributing to regulation of neurotransmitter release and synaptic plasticity.

RGS20 influences several G-protein mediated pathways such as the cAMP signaling pathway and the MAPK pathway. Inhibits signal transduction by increasing the GTPase activity of G protein alpha subunits thereby driving them into their inactive GDP-bound form. Binds selectively to G(z)-alpha and G(alpha)-i2 subunits, accelerates their GTPase activity and regulates their signaling activities. The G(z)-alpha activity is inhibited by the phosphorylation and palmitoylation of the G-protein. Negatively regulates mu-opioid receptor-mediated activation of the G-proteins (By similarity).
