The RHAG protein also known as the Rh-associated glycoprotein or Rh family C glycoprotein has a molecular mass of approximately 50000 Daltons. It is present mainly on erythrocyte membranes and works as a part of the Rh blood group system. RHAG acts as an ammonium transporter enabling the movement of ammonia and possibly CO2 across the cell membrane though exact specifics of its mechanisms are not completely clear.
Biological function summary

The RHAG protein is part of the Rh complex within the erythrocyte membrane which also includes RhD and RhCE proteins. This complex contributes to the structural integrity of the red cell membrane and plays an important role in maintaining the biconcave shape of the erythrocytes which is necessary for optimal oxygen exchange and gas transport. RHAG's function is essential for normal erythrocyte operation and overall cellular homeostasis.

Pathways

RHAG participates in the ammonia transport pathway and is linked to the Rh protein complex pathway. It interacts closely with proteins like RhD and RhCE working together to facilitate membrane stability and function in red blood cells. These interactions within the pathways emphasize RHAG's role in gas transport-related functions and metabolic waste management in the bloodstream.

Defects in RHAG expression or function lead to conditions like Rh-null syndrome and hereditary stomatocytosis. This syndrome characterized by a lack of all Rh antigens on red blood cells often associates with hemolytic anemia due to the compromised integrity of the erythrocyte membrane. Mutations in RHAG also relate to interactions with other proteins like SLC4A1 another important protein involved in red blood cell homeostasis enhancing the complexity and clinical significance of the RHAG protein in health and disease contexts.
