Supplementary information

RHBDD2 also known as rhomboid domain containing 2 is a serine protease that plays significant roles in intramembrane proteolysis. It has a molecular mass of approximately 44 kDa. This protein resides in the membranes of cellular endoplasmic reticulum and Golgi apparatus and it is expressed in various tissues including breast prostate and liver. RHBDD2 exhibits gene expression in both normal and cancerous cells suggesting its importance in physiological and pathological conditions.
Biological function summary

RHBDD2 participates in protein processing and quality control within the cell. It functions in the regulated intramembrane proteolysis of substrates impacting signaling pathways related to cell growth and apoptosis. Though RHBDD2 does not belong to a specific complex its proteolytic activity allows it to cleave and modulate activity of other membrane-associated proteins affecting their function and stability. This role positions RHBDD2 as an influential factor in determining protein fate within the cell.

Pathways

RHBDD2 embeds itself in the network of the ER-associated degradation (ERAD) pathway and the apoptosis signaling pathway. In the ERAD pathway RHBDD2 helps degrade misfolded proteins maintaining cellular homeostasis. In the apoptosis signaling pathway RHBDD2 indirectly influences through cleavage of proteins such as BCL2 family members integrating itself into pro-apoptotic and anti-apoptotic signals. Its connection with proteins like ubiquitin ligases in these pathways shows operation in cellular protein management.

RHBDD2 has associations with cancer and neurodegenerative disorders. Its overexpression in breast and prostate cancer implies a link with tumor growth and progression. In neurodegenerative conditions RHBDD2 expression can alter the processing of proteins like amyloid precursor protein (APP) impacting pathologies like Alzheimer's disease. These connections suggest that RHBDD2 along with its interacting molecules plays roles in cell proliferation and neurodegeneration processes.
