RHOBTB1 also known as Rho-related BTB domain-containing protein 1 is a member of the Rho GTPase family. It has a molecular mass of approximately 82 kDa. This protein is mainly expressed in the brain and testis with lower levels found in other tissues. RHOBTB1 functions by acting as a molecular switch that cycles between an active GTP-bound state and an inactive GDP-bound state which regulates various cellular processes.
Biological function summary

RHOBTB1 is involved in the regulation of cytoskeletal dynamics which affects cell shape motility and possibly cell division. It is also implicated in intracellular signaling processes that control cell morphology. RHOBTB1 does not function alone; it interacts with other proteins possibly forming part of larger protein complexes that further expand its role in cellular activities.

Pathways

RHOBTB1 plays a role in signaling pathways that include the Rho GTPase cycle and cellular processes related to cytoskeletal reorganization. In these pathways RHOBTB1 interacts with proteins such as CDC42 and RAC1 which are essential for actin filament polymerization and cellular movement. These interactions help facilitate normal cellular dynamics and responses to extracellular signals.

Mutations or dysregulation of RHOBTB1 have been linked to neurological conditions and certain cancers. For example RHOBTB1 abnormalities have been associated with an increased risk of glioblastoma where it might interact with tumor suppressor proteins like p53. Additionally RHOBTB1's role in vacuole formation has implications in lysosomal storage diseases connecting it to other lysosomal proteins involved in these processes.
