Immunohistochemical analysis of paraffin-embedded Human tonsil tissue labeling RIAM with ab247579 at 1/2000 (0.449 ug/ml) dilution.

The section was incubated with ab247579 for 30 mins at room temperature. Counterstained with hematoxylin. The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins.

RIAM also known as Rap1-GTP-interacting adaptor molecule is a protein involved in intracellular signaling. RIAM has a molecular weight of approximately 76 kDa. It is expressed in various tissues with a significant presence in hematopoietic cells and endothelial cells. This protein plays a part in connecting cell membrane molecules with actin cytoskeleton therefore promoting cellular adhesion.
Biological function summary

RIAM is essential for the integrin-mediated adhesion processes. It functions as a critical adaptor forming complexes with talin and other signaling proteins which facilitate the activation of integrins. Through these actions RIAM contributes to cell migration immune responses and platelet aggregation. Its interactions influence cellular adhesion and communication pivotal for maintaining tissue structure and response to injuries.

Pathways

RIAM is a vital component in the signaling pathways involving the small GTPase Rap1 which regulates integrins and their effect on cell adhesion. Particularly RIAM serves as a bridge between Rap1 and talin facilitating integrin activation. This process plays an essential role in the focal adhesion pathway where proteins like Kindlin also collaborate to modulate cellular adhesion and movement impacting various physiological processes.

RIAM has connections with immune dysfunction and cardiovascular diseases. For example abnormalities in RIAM-mediated integrin activation can lead to improper immune cell trafficking and related immunodeficiencies. In cardiovascular contexts defective RIAM activity might impair endothelial cell function and contribute to vascular diseases. In these scenarios proteins like talin are closely associated with RIAM often sharing roles in the pathogenesis of these disorders.
