Lane 1 : Whole cell lysate from Jurkat cells (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) prepared using NETN lysis buffer. ab194941 was used for IP at 6 μg per reaction, and at 1 μg/ml for blotting the immunoprecipitate.

Lane 2 : Control IgG.

Detection : Chemiluminescence with an exposure time of 30 seconds.

RIC8A also known as Synembryn-A is a guanine nucleotide exchange factor that plays an important mechanical role in cellular signaling. It catalyzes the exchange of GDP for GTP on G protein alpha subunits activating them. RIC8A weighs approximately 60 kDa and is mainly cytoplasmic. Researchers have identified its expression in several tissues such as the brain heart and lungs indicating a wide pattern of activity in the body.
Biological function summary

RIC8A is involved in intracellular signaling and regulation of G-protein coupled receptor (GPCR) pathways. It does not directly form part of a stable complex but interacts transiently with heterotrimeric G proteins during activation. Chaperone that specifically binds and folds nascent G alpha proteins prior to G protein heterotrimer formation, promoting their stability and activity : folds GNAI1, GNAO1, GNA13 and GNAQ (By similarity). Does not fold G(s) G-alpha proteins GNAS nor GNAL (By similarity). Also acts as a guanine nucleotide exchange factor (GEF) for G alpha proteins by stimulating exchange of bound GDP for free GTP (By similarity). Involved in regulation of microtubule pulling forces during mitotic movement of chromosomes by stimulating G(i)-alpha protein (GNAI1), possibly leading to release G(i)-alpha-GTP and NuMA proteins from the NuMA-GPSM2-G(i)-alpha-GDP complex (By similarity).
