RIC8B also known as Resistance to Inhibitors of Cholinesterase 8 homolog B is a guanine nucleotide exchange factor that interacts with G protein alpha subunits. It has a molecular weight of approximately 60 kDa. This protein is expressed in many tissues with high levels reported in areas of the brain such as the cerebral cortex and cerebellum. RIC8B plays an important role in modulating signal transduction by facilitating the replacement of bound GDP with GTP on G protein alpha subunits which is an essential step in signal transducing processes.
RIC8B acts as a regulatory element in G protein signaling. It is not a permanent part of a protein complex but associates transiently with G alpha subunits to promote their active form. These interactions are critical for downstream signaling cascades that influence cellular responses to external stimuli. RIC8B's role in conjunction with G protein-coupled receptors (GPCRs) affects how cells perceive and respond to a wide range of signals including hormones neurotransmitters and other signaling molecules.
RIC8B influences multiple signaling pathways notably the GPCR and olfactory signaling pathways. In GPCR signaling RIC8B interacts with G protein alpha subunits such as GNAI and GNAS to enhance the signal transduction process. Through these pathways RIC8B affects various physiological processes like olfaction and balance. Additionally its regulation of G alpha proteins integrates with other proteins such as GPCR kinases to modulate receptor desensitization and signaling duration.
RIC8B has connections to neurodegenerative disorders like Alzheimer's and bipolar disorder. Disruption of its function can impair neurotransmitter signaling thereby influencing disease pathways. RIC8B's interactions with proteins like ApoE in Alzheimer's and with neurotransmitter systems in mood disorders suggest roles in disease mechanisms.
