RIN3 also known as Ras and Rab interactor 3 is a member of the RIN family of proteins. This protein has a mass of approximately 100 kDa. RIN3 functions as a GDP-GTP exchange factor which is important for the regulation of endocytosis and actin cytoskeleton organization. It primarily helps in activating small GTPase proteins particularly RAB5 and RAB31. RIN3 expression is found in various tissues with higher levels noted in bone and immune-related tissues.
Biological function summary

This protein influences several important cellular processes including intracellular trafficking and signal transduction. It does not form large complexes itself but it interacts with other proteins like BIN2 which facilitates endocytic vesicle formation and cellular signaling. By these interactions RIN3 contributes to actin cytoskeleton rearrangement and membrane ruffling impacting the cellular uptake and movement.

Pathways

Significant involvement occurs in the endocytic pathway and actin cytoskeleton regulatory pathway. Within the endocytic pathway RIN3 interacts with RAB5 and other related GTPases such as RAB31 orchestrating early endosome dynamics. In the actin cytoskeleton pathway interactions with BIN2 play a role in actin filament modulation which influences cell shape and motility.

RIN3 has been linked to Paget's Disease of Bone and some forms of immunodeficiency. This protein's dysfunction in these conditions relates to its role in bone resorption and immune cell function. Additionally it interacts with SQSTM1 which is associated with bone metabolism and STK11 which relates to immune function disorders. Understanding these interactions and pathways provides insight into RIN3's involvement in disease mechanisms.
