Western blot was performed on the immunoprecipitate using ab180170 at 1/2000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/10000 dilution.

Lanes 1 - 4 : Merged signal (red and green). Green - ab180170 observed at 52 kDa. Red - loading control, ab8245, observed at 37 kDa.

ab180170 was shown to specifically react with RING1 when RING1 knockout samples were used. Wild-type and RING1 knockout samples were subjected to SDS-PAGE. ab180170 and ab8245 (loading control to GAPDH) were diluted 1/1000 and 1/10000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

RING1 also known as Ring Finger Protein 1 or RNF1 is an E3 ubiquitin-protein ligase with a molecular mass of approximately 42 kDa. It is part of the polycomb group proteins known for regulating gene expression. RING1 is expressed ubiquitously in various tissues where it functions as an important regulator of protein degradation by tagging substrates with ubiquitin for proteasomal degradation. This activity plays a critical role in maintaining cellular homeostasis by controlling the abundance of regulatory proteins.
Biological function summary

Along with modulating protein turnover RING1 participates as a fundamental component of the Polycomb Repressive Complex 1 (PRC1). This complex is imperative in chromatin remodeling and transcriptional repression influencing epigenetic gene silencing. RING1 works in tandem with other proteins such as BMI1 facilitating histone ubiquitination and altering chromatin structure which impacts gene expression across different cell types and developmental stages.

Pathways

The ubiquitin-proteasome system and the epigenetic regulation pathway involve RING1. Within the ubiquitin-proteasome system RING1 interacts with other E3 ligases and ubiquitin-conjugating enzymes governing the fate of its protein substrates. In the epigenetic regulation pathway RING1's role in PRC1 places it alongside proteins like EZH2 contributing to the repression of genes involved in differentiation and development thereby ensuring proper cell identity maintenance.

Mutations or misregulation of RING1 have links to several conditions including cancers and neurodegenerative disorders. In cancer the disruption of normal RING1 functions can lead to unregulated cell proliferation due to improper gene silencing. For neurodegenerative disorders associations with proteins such as ubiquitin and its regulatory roles may contribute to disease pathogenesis by enabling the accumulation of misfolded proteins further highlighting its significance in cellular health and disease.

Constitutes one of the E3 ubiquitin-protein ligases that mediate monoubiquitination of 'Lys-119' of histone H2A, thereby playing a central role in histone code and gene regulation. H2A 'Lys-119' ubiquitination gives a specific tag for epigenetic transcriptional repression and participates in X chromosome inactivation of female mammals. Essential component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones, rendering chromatin heritably changed in its expressibility. Compared to RNF2/RING2, it does not have the main E3 ubiquitin ligase activity on histone H2A, and it may rather act as a modulator of RNF2/RING2 activity.
