RNA polymerase beta was immunoprecipitated from 1mg of E.coli whole cell lysates with ab191598 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab191598 at 1/1000 dilution. Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/1500.

Lane 1 : E.coli whole cell lysates 10ug (Input). Lane 2 : ab191598 IP in E.coli whole cell lysates. Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab191598 in E.coli whole cell lysates.

Blocking and dilution buffer and concentration : 5% NFDM/TBST.

Exposure time : 5 seconds.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab191598).

RNA polymerase beta also known as RpoB or DNA-directed RNA polymerase subunit beta is a critical enzyme in the transcription process. This protein has a molecular mass of approximately 150 kDa. RpoB plays a role by catalyzing the synthesis of RNA from a DNA template an essential step in gene expression. It is widely expressed in bacteria mitochondria chloroplasts and also in the nuclei of eukaryotic cells. RpoB functions as a fundamental part of the core RNA polymerase complex which also includes other subunits such as alpha beta' and omega.
Biological function summary

RpoB interacts with multiple subunits to form the RNA polymerase holoenzyme integrating into cellular machinery to initiate transcription. This enzyme participates in synthesizing mRNA rRNA and tRNA all of which are important for protein synthesis. The holoenzyme formation aligns with promoter regions on DNA enabling precise transcription initiation. Without the proper function of RpoB and its associated subunits transcription efficiency and fidelity could be compromised impacting cell viability.

RNA polymerase beta is integral to transcriptional regulation pathways and genetic response mechanisms within the cell. It plays a role in the bacterial transcription pathway directly interacting with sigma factors and other transcription regulators to manage gene expression in response to environmental changes. DNA-dependent RNA polymerase (RNAP) catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates.. Resistance to the antibiotics salinamide A, salinamide B, rifampicin, streptolydigin, CBR703, myxopyronin, and lipiarmycin can result from mutations in this protein.. Part of the processive rRNA transcription and antitermination complex (rrnTAC). The complex forms an RNA-chaperone ring around the RNA exit tunnel of RNAP. It supports rapid transcription and antitermination of rRNA operons, cotranscriptional rRNA folding, and annealing of distal rRNA regions to allow correct ribosome biogenesis.
