Supplementary information

RND2 also known as Rhonadin-2 is a small GTP-binding protein with a molecular weight of approximately 24 kDa. It belongs to the Rho family of GTPases and functions mechanically as a regulator of the actin cytoskeleton. This protein controls cellular processes such as migration and cytoskeletal organization through its ability to cycle between an active GTP-bound state and an inactive GDP-bound state. RND2 is expressed in various tissues but shows significant presence in the neural tissues where it plays an important role in neuronal morphology.
Biological function summary

This protein plays a significant role in neuronal development and differentiation. It is not typically a part of other protein complexes but directly interacts with effector proteins involved in actin dynamics. RND2 modulates processes such as axonal guidance and dendritic spine formation contributing to nerve cell communication and network formation during brain development. Its ability to control actin polymerization helps in shaping the structural integrity of neurons which is essential for their proper functioning.

Pathways

RND2 influences cellular actions by integrating into the actin cytoskeleton signaling pathway and the neurotrophin signaling pathway. The actin cytoskeleton pathway involves changes in cell shape and movement through interactions with proteins like ROCK2 and cofilin which RND2 regulates. Neurotrophin pathways affect neuronal survival and growth where RND2 acts alongside proteins like NTRK2 impacting the development and maintenance of the neuronal system.

RND2 has implications in neurodevelopmental disorders and cancer. Mutations or dysregulation of this protein might contribute to conditions like intellectual disability due to its critical role in neuronal development. Moreover aberrant expression of RND2 links to cancer metastasis as its regulation of cell movement and invasion can drive tumor progression. In cancer RND2 interacts with proteins like Rac1 affecting pathways that control tumor cell migration and invasion.
